ドラマの舞台となるのは、海上保安庁に新設された「ＤＣＵ」という水中事件や事故の捜査を行うスペシャリスト集団。ＤＣＵはDeep Crime Unit（潜水特殊捜査隊）の略称で、彼らの使命は、従来の海上水域だけでなく警察の捜査では困難な「危険極まりない日本全国の河川や湖」など、あらゆる水中に潜り隠された証拠を探し事件を解決する。“水中未解決事件を解明すること”が、彼らのミッションだ。
ＤＣＵが“水”にまつわる事件に遭遇して人の命と尊さを厳粛に扱う一方で、彼らの人間模様をしっかりと描いていく。個性豊かなメンバーたちの人間ドラマを中心に描きながら、さまざまな水中事件や救助、外部からの侵入対策など難事件に立ち向かい、「水中」にある謎や事件をスピード感と爽快感をもって解明していく。
