Having received a letter saying "We are being attacked by the youkai Mannendake," Kitaro travels to a bamboo grove deep in the mountains. The owners of the bamboo grove, a married couple, Masahiko and Kiyomi, Kubota from the tourism association, and Ratman are waiting for him. They wanted to turn the bamboo grove into a tourist spot, but they say that the former owner of the grove, Daikichi, as well as others, have gone missing. As soon as Kitaro steps foot in the grove, he senses youkai energy. A warning to humans is written in blood in the grove, but the study in Daikichi's mansion within the grove is full of bamboo leaves... Suddenly, Masahiko falls silent. When they turn to look, Masahiko has been turned to bamboo...!?

「妖怪・万年竹に襲われる」という手紙を受け取った鬼太郎は、山深くの竹薮までやってきた。そこには、竹薮の持ち主である雅彦と清美の夫妻、観光協会の久保田、ねずみ男が待ち構えていた。彼らは竹薮を観光地にしようと考えたが、竹薮の元主である大吉をはじめ次々と行方不明者が出ているのだという。竹薮に足を踏み入れた鬼太郎は、途端に妖気を感じる。藪の中には血塗りの文字で人間への警告文が、奥にある大吉の屋敷の中の書斎は竹の葉だらけ…。突如、怯え続けていた雅彦が静かになった。見ると、雅彦の全身は竹になっていて…！？
