Gary Navarro is arrested for the second attempt to assassinate Dennis Ryland. Knowing she is the key to her mother's survival, Isabelle attempts to kill herself by jumping off the 4400 center building only to survive without a scratch. Matthew Ross expresses his displeasure at her actions and tells her of the destiny she is to face. Isabelle is troubled about what Matthew told her, but after a discussion with Shawn she decides it's her choice with what she does with her life. Matthew gives Isabelle a substance that can kill her if it's injected.
Le groupe Nova annonce le début d'une nouvelle ère sous forme d'ultimatum aux autorités. Le NTAC s'attend à un acte terroriste de grande ampleur. Pendant ce temps, Gary Navarro est arrêté pour la tentative d'assassinat de Ryland et remis aux mains de la NSA. Tom et Diana, dépossédés de ce témoin, s'inquiètent maintenant pour les 4400 dont les pouvoirs sont décuplés depuis l'arrêt de l'inhibiteur. De retour au centre, Matthew contraint Isabelle à prendre son destin en main alors que sa mère est condamnée à mourir. Se sentant responsable de cette situation, elle tente de mettre fin à ses jours. Enfin, Shawn rompt ses engagements auprès du groupe Nova...
Gary Navarro viene arrestato per aver tentato di assassinare una seconda volta Ryland. Isabelle, credendo di salvare la vita a sua madre uccidendosi, si getta dal tetto del centro 4400, ma si rialza senza un graffio. Matthew Ross esprime il suo disappunto nei confronti di Isabelle per ciò che ha tentato di fare le parla dell’importanza della sua esistenza per il futuro dell’umanità. Isabelle, è preoccupata per ciò che le ha detto Matthew, ma dopo aver parlato con Shawn decide di essere lei l’artefice delle sue scelte e non il destino. Matthew dà ad Isabelle una sostanza che iniettata è in grado di ucciderla.
Isabelle intentará suicidarse, aunque, afortunadamente, sobrevive sin tener ningún rasguño.
