シェイクスピアの「マクベス」を日本の戦国時代に置き換え描いた、戦国武将の一大悲劇。謀叛を起こした敵を討ち城主の危機を救った鷲津武時は、帰城途中に出会った老婆から不思議な予言を聞く。その予言通りに大将に任ぜられると、今度は妻にそそのかされて主を殺害、自ら城主の地位に着く。黒澤監督は、欲望に刈られた魂が繰り返す殺戮と狂気を、能の様式美に乗せて見事に描いていく。三船＝マクベスが無数の矢に曝されるラストシーンは圧巻。

O general Washizu e o general Miki perdem-se na floresta e encontram uma bruxa que prevê que Washizu será rei e que será sucedido pelos herdeiros de Miki. É então que Washizu mata o seu senhor, Kuniharu Tsuzuki, e Miki. A bruxa prevê agora que ele estará a salvo enquanto a floresta não se puser em movimento. Mas o filho de Miki ataca o castelo de Washizu, usando as árvores da floresta como camuflagem. O filho de Washizu nasce morto, a sua mulher enlouquece e ele é traído pelos seus homens. Adaptação livre de Macbeth de William Shakespeare, "O Trono de Sangue" introduz pequenas diferenças em relação à peça (uma só bruxa, ao invés de três) e acrescenta novas cenas (quando a floresta se põe em movimento, os pássaros fogem e invadem o castelo).

Generals Miki and Washizu are Samurai commanders and friends under a local lord, Lord Tsuzuki, who reigns in the castle of the Spider's Web Forest. After defeating the lord's enemies in battle, they return to Tsuzuki's castle. On their way through the thick forest surrounding the castle, they meet a spirit, who foretells their future. The spirit tells them that today Washizu will be named Lord of the Northern Garrison and Miki will become commander of the first fortress. She then foretells that Washizu will eventually become Lord of Spider's Web Castle, and finally she tells Miki that his son will also become lord of the castle. When the two return to Tsuzuki's estate, he rewards them with exactly what the spirit had predicted. As Washizu discusses this with Asaji, his wife, she manipulates him into making the second part of the prophecy come true by murdering Tsuzuki when he visits.

Japão. Yoshiteru Miki (Akira Kubo) e Taketori Washizu (Toshirô Mifune) são os comandantes do primeiro e do segundo castelo de um reino local, cuja sede fica no Castelo das Teias de Aranha. Após defenderem seu senhor em batalha, eles estão retornando para casa quando encontram um espírito que prediz o futuro de ambos. Ele diz que Washizu em breve assumirá o trono e que o filho de Miki, Yoshaki (Minoru Chiaki), o sucederá. Ao retornar para casa, Washizu comenta a predição com a esposa, lady Asaji (Isuzu Yamada). Ela acredita no que o espírito disse e incentiva o marido a agir quando o atual rei chega em seu castelo, para passar a noite.
